Sox2 is a transcription factor whose activity marks and maintains embryonic stem cells, and is also required for reprogramming somatic cells into induced pluripotent stem cells (IPC). We will show that Sox2 expression also marks and maintains adult stem cells in the bone lineage through the activation of a complex series of transcriptional and posttranscriptional events which are necessary of their self-renewal. In osteosarcomas, the most common bone tumor, Sox2 plays a similar role by maintaining the cancer stem cell population through the activation of a network of transcriptional and signaling events that is indispensable for tumorigenesis.
